MT extension during growth cone turning evoked by beads coated with the Ig superfamily cell adhesion protein apCAM occurred in concert with attenuation of retrograde F-actin flow. Interestingly, blocking MT dynamics resulted in inhibition of growth cone steering by uncoupling Src-dependent adhesion receptor-cytoskeletal interactions. The authors suggest that pioneer MTs are involved in local activation of Src at adhesion sites.

Interestingly, PlexinA4 appears not to be necessary for Slit2 repulsive steering events at the central nervous system midline. Thus, the study provides a potential mechanistic explanation for previous findings reporting that Slit2 repels some axons but promotes growth (branching) of others.

Strikingly, projection neurons in the hippocampus and cerebral cortex of kif2A mutants show aberrant axonal branching due to overextension of collateral branches. Biochemical studies coupled with live imaging of MTs in kif2A mutants provide strong evidence that KIF2A functions as a 'catastrophe promoting' MT destabilizing factor in vitro and in vivo. This suggests that destabilization of MT ends by KIF2A is a potential mechanism to suppress supernumerary collateral axon branches in vivo.

Overexpression of orbit/MAST enhances the Abl GOF phenotype. Through phenotypic characterization, genetic interactions, and epistasis experiments the authors provide strong evidence that orbit-MAST functions downstream of Abl in the Slit-Robo repellent pathway. In a parallel approach, the authors express GFP-CLASP in Xenopus motoneurons and show that CLASP has 'tip tracking' activity and binds to the plus end of pioneer MTs. CLASP not only binds to MT plus ends but also reduces MT advance and growth cone translocation. When combined with previous findings, these elegant studies provide strong evidence that Abl controls F-actin and MT dynamics downstream of guidance receptors.

NGF-mediated activation of PI3-kinase leads to local inactivation of GSK3β. The authors go on to show that these two specially coupled kinases control axon growth by regulating the microtubule plus end binding protein APC. Imaging studies directly showed that APC is localized to the plus end of MTs in growth cones. Mutagenesis studies revealed that spatial localization of APC to MT tips is necessary for NGF induced axon growth. These important findings link NGF signaling to the conserved signaling pathway known to regulate MT dynamics in cell polarization.

In parallel, activation of Rac1 promoted lamellipodial actin polymerization and Pak-dependent retrograde flow. Taken together, the results show that Pak-activity is required for Rac1-induced changes in actin and MT organization and dynamics.

These observations link two important signaling pathways implicated in cell polarity, the Cdc42-Par proteins and the GSK3-β-catenin-APC complex.
