    • The morphogen sonic hedgehog is an axonal chemoattractant that collaborates with netrin-1 in midline axon guidance
    • Using conditional knockout mice and in vitro assays, the authors show that Shh functions as a chemoattractant for commissural axons, thereby cooperating with Netrin-1 in the guidance of these axons to the floor plate. This study provides the first evidence to suggest that a morphogen gradient, initially used for specifying cell fates, is re-used to guide axons.

    • A role for BMP heterodimers in roof plate-mediated repulsion of commissural axons
    • Using knockout mutant mice and in vitro assays, the authors provide evidence that BMP7 acts as a chemorepellent for commissural axons, probably by forming heterodimers with GDF7, another member of the TGF-β family that is secreted by the roof plate. Together with the study of Shh [1], this work suggests that morphogen gradients may be used for axon guidance.

    • Wnt-mediated axon guidance via the Drosophila derailed receptor
    • The authors show that Wnt5, one of the seven Drosophila members of the Wnt family, acts as a chemorepellent to guide axons through the anterior commissure, one of the two principal axon tracts that cross the midline. Wnt5 is shown to bind and to require a new Wnt receptor called Derailed, an atypical receptor tyrosine kinase.

    • Anterior-posterior guidance of commissural axons by Wnt-Frizzled signaling
    • The authors show that secreted Wnt4 can reorient commissural axons in spinal cord explants and that the Wnt4 gene is transcribed in an increasing posterior-to-anterior gradient within the floor plate. With the additional finding that commissural axons fail to project normally in Frizzled3 mutant mice, this paper provides evidence for a Wnt family member acting as a chemoattractant for commissural axons once they have crossed the floor plate.
