These observations suggest that NO signaling in cells is selectively targeted to cGMP responses, rather than inhibition of cytochrome oxidase, reflecting the differential sensitivity of guanylyl cyclase, which is nearly two orders of magnitude more sensitive to NO compared to cytochrome oxidase under normoxic conditions, and the maximum concentrations of NO achieved in cells, which is well below the binding constant for cytochrome oxidase under normoxic conditions. Indeed, these data argue for a specific role for mitochondrial NOS in regulating cytochrome oxidase, ATP generation, and oxygen consumption.
