This paper describes the characterisation of a novel cGMP signalling pathway in Dictyostelium and analyses its role in chemotaxis. Cells with a reduction in phosphodiesterase activity showed enhanced chemotaxis, whereas cells that lack cGMP binding proteins GbpC and GbpD show severely impaired chemotaxis.

Cells first become very motile and show a highly polarised morphology. This is followed by cell depolarisation and vacuolisation.

SadA is the first substrate-adhesion receptor to be identified in Dictyostelium. This protein is only expressed in growing cells. Cells lacking sadA show impaired cytokinesis, aberrant F-actin organisation and a phagocytosis defect.

Cell 2003, 114:559-572. IP7/IP8 levels are raised during chemotactic stimulation. Loss of IP7/IP8 by deletion of IP6 kinase leads to early aggregation and hypersensitivity to cAMP. IP7 binds to PH domain containing proteins and competes with their binding to PIP3.
