    • Intestinal stem cells protect their genome by selective segregation of template DNA strands
    • 3H -thymidine during mice development, or into adult animals that have been irradiated. After several cell generations, the label is only retained by stem cells.     • Investigating stem cells in human colon by using methylation patterns
    • The authors study CpG methylation tags of three neutral loci in purified individual crypts from human colon. Since crypt cells accumulate random methylation errors, they are able to quantitatively compare phylogenies within the same crypts and between different crypts. Applying mathematical simulations, they conclude that the best model to explain their data are the existence of crypt niches where a small number of stem cells can divide symmetrically or asymmetrically following a stochastic pattern.

    • This study revealed the role of Wnt signalling in the control of the proliferation/differentiation switch in adult intestinal homeostasis; it also demonstrates the presence of nuclear β-catenin in crypts. It describes part of the genetic programme controlled by the Wnt signalling in colorectal cancer cells where the activity of the β-catenin-TCF complex was inhibited by the inducible expression of dominant-negative TCF constructs.

    • This paper describes how β-catenin-TCF activity inversely controls the expression of the receptor tyrosine kinase EphB2 and EphB3 and their ligands ephrin-B1 and -B2 along the crypt axis. The analysis of mice deficient for these receptors showed that they allocate different cell types in the intestinal epithelium. This includes the boundary between the proliferative and differentiated compartment in neonatal mice, and the localisation of Paneth cells and undifferentiated precursors on the adult crypts.
