    • Do human cancers express shared protective antigens? Or the necessity of remembrance of things past

Attention is drawn to the fact that for the identification of tumor antigens rejection assays are used in animal models, whereas in vitro generated cytotoxic T lymphocytes have been used as the probes for the identification of shared tumor antigens in humans. The controversy between the finding of unique antigens in animal models and shared antigens in humans by these methods is discussed

    • The SSX2 gene, which is involved in the t(X, 18) translocation of synovial sarcomas, codes for the human tumor antigen HOM-Mel-40
    • of outstanding interest. HOM-MEL-40 antigen detected in a melanoma library is encoded by the SSX-2 gene, and is the first cancer/testis antigen (i.e. a testicular antigen aberrantly expressed in human neoplasms in a lineage independent pattern) identified by SEREX. SSX2 is silenced in normal tissues, except for the testis, but is expressed in 8 out of 16 melanomas, as well as in 7 out of 36 breast cancers, 9 out of 35 colorectal carcinomas and in a lower frequency in prostate cancer and glioma. 10% of melanoma patients raise IgG responses to SSX2, SSX2 is known to participate in the t(X;18) translocation frequently found in synovial sarcoma.
