The mechanism by which PU.1 deletion inhibits lymphoid lineage specification was determined in this study to be at least partially owing to a failure of interleukin-7 receptor expression in the absence of PU.1. Coupled with the observation that graded expression of PU.1 regulates myeloid versus lymphoid lineage choices [5], this study establishes the mechanism of early lymphoid lineage commitment.

neg thymocyte development has no effect on the subsequent expression of CD4 and CD8 or on the maturation and survival of thymocytes. The specific role of the thymus in directing T lineage development has been questioned for decades. By expressing the Notch ligand Delta-like-1 in the OP9 cell line, these workers have succeeded in establishing an in vitro model of T cell commitment and differentiation that is independent of primary thymic stromal cells. The OP9 cell line, which was derived from the op/op mutant mouse and thus does not produce macrophage colony-stimulating factor, is a key feature of the model system.

This study demonstrates that interference of Notch-1 signaling by Deltex1 in lymphoid progenitor cells suppresses T-lineage development while promoting B lineage development. The mechanism by which Deltex1 mediates this effect involves displacement of transcriptional coactivators from the nuclear complex of Notch-1 with other transcription factors. Thus, Notch-1 activity can be regulated at multiple levels in developing lymphocytes.
