Mutant PU.1 cDNAs were constructed using PCR as described [R. C. Fisher et al., Mol. Cell. Biol. 18, 4347 (1998)] except for the 5′ external primer (5′-CGGCGGCCGCTTACAGGCGTGCAAAATGGAAGGGT-3′) and 3′ external primer (5′-CCGGGCCCTCAGTGGGGCGGGAGGCGCCG-3′) and verified by DNA sequencing. All PU.1 mutant forms contained a 5′ HA-tag sequence. All constructs were blunt-end-ligated into the Hpa I site of MSCV-EGFP. Packaging cell lines were constructed as described (10) and expressed similar concentrations of the various PU.1 derivatives.
