    • Regulation of dendritic spine morphology and synaptic function by Shank and Homer
    • In this paper, the authors transfected primary hippocampal neurons with Shank and or Homer, respectively. Overexpression of these scaffolding proteins increased spine size and number, and promoted the accumulation of associated scaffolding and signaling proteins. Surprisingly, postsynaptic expression enhanced presynaptic functions, as mEPSC frequencies were significantly higher for transfected neurons. These observations suggest that scaffolding proteins may play significant roles in regulating synaptic formation and plasticity.

    • Stargazin mediates synaptic targeting of AMPA receptors by two distinct mechanisms
    • Stargazin, a transmembrane protein, is required for the surface and synaptic expression of AMPA receptors in cerebellar granule neurons. Expression of a truncated version of Stargazin lacking the PDZ-binding motif disrupts the synaptic accumulation of AMPA receptors. This study suggests this family of proteins may play an essential role in regulating the trafficking and synaptic retention of glutamate receptors.

    • Synaptic strength regulated by palmitate cycling on PSD-95
    • Here, these authors use 2-bromopalmitate to acutely block palmitoylation in heterologous cells and primary hippocampal cultures. Application of the drug disperses synaptic PSD95 and AMPA receptors but not NMDA receptors, and reduces miniature excitatory postsynaptic current amplitude and frequency. The declustering of PSD95 by 2-bromopalmitate is regulated by synaptic activity, as glutamate receptor antagonists diminish this effect and decrease the palmitate turnover on PSD95. This work suggests that palmitate cycling on PSD95 can regulate aspects of synaptic plasticity.
