    • Schwarz-Romond T., Asbrand C., Bakkers J., Kühl M., Schaeffer H.-J., Huelsken J., Behrens J., Hammerschmidt M., Birchmeier W. The ankyrin repeat protein Diversin recruits Casein kinase Iε to the β-catenin degradation complex and acts in both canonical Wnt and Wnt/JNK signaling. Genes Dev. 16:2002;2073-2084 The authors describe the identification and characterization of a novel member of the Wnt signaling cascade: Diversin. This molecule acts as a molecular switch between the canonical Wnt pathway and the PCP pathway. The Wnt signal mediated by the canonical pathway is suppressed and signaling via JNK is stimulated. This mode of action is achieved by Diversin's interaction with CK1e and Axin/Conductin.

    • Mao J., Wang J., Liu B., Pan W., Farr G.H., Flynn C., Yuan H., Takada S., Kimelman D., Li L., Wu D. Low-density receptor related protein 5 binds to axin and regulates the canonical Wnt signalling pathway. Mol. Cell. 7:2001;801-809 The authors show in this paper that axin, a scaffold protein in the Wnt signaling cascade, is able to interact directly with the intracellular domain of the membrane receptor LRP5. The addition of Wnt causes the translocation of axin to the membrane and enhances the interaction between axin and LRP5. The binding of axin to LRP5 induces the degradation of axin, which leads to LEF-1 mediated transcriptional activation.

Cell. 108:2002;837-847 This excellent paper describes the role of CK1 in β-catenin regulation. In the absence of Wnt, β-catenin is phosphorylated at residue Ser45 by CK1, which subsequently serves as the priming site for phosphorylation by GSK3β. This leads to degradation of β-catenin.

Genes Dev. 16:2002;1066-1076 The authors show that a complex of axin and CKI induces β-catenin phosphorylation at residue Ser45. This CKI phosphorylation of β-catenin creates a priming site for GSK-3β and is both necessary and sufficient to initiate the degradation cascade of β-catenin.

Wnt/Wingless signaling requires BCL9/Legless-mediated recruitment of Pygopus to the nuclear β-catenin-TCF complex. Cell. 109:2002;47-60 Using strong genetic studies in Drosophila, this paper reports the identification of two novel components of the Wnt signaling cascade, legless and pygopus, which are both necessary for Wnt signal transduction at the level of nuclear β-catenin. In addition, they provide genetic and molecular evidence that these proteins exert their function by physically linking Pygo via Legless to β-catenin.
