The addition of Wnt causes the translocation of axin to the membrane and enhances the interaction between axin and LRP5. The binding of axin to LRP5 induces the degradation of axin, which leads to LEF-1 mediated transcriptional activation.

Cell. 109:2002;47-60 Using strong genetic studies in Drosophila, this paper reports the identification of two novel components of the Wnt signaling cascade, legless and pygopus, which are both necessary for Wnt signal transduction at the level of nuclear β-catenin. In addition, they provide genetic and molecular evidence that these proteins exert their function by physically linking Pygo via Legless to β-catenin.
