    • Normal limb development in conditional mutants of Fgf4
    • Using a conditional null allele of Fgf-4 to bypass early embryonic lethality, the authors provide evidence that Fgf-4 signaling is not required for maintenance of Shh expression. In addition, the limb skeleton appears to develop normally. Together, these data suggest that multiple FGF signals from the AER maintain Shh expression.

    • Dispatched, a novel sterol-sensing domain protein dedicated to the release of cholesterol-modified hedgehog from signaling cells
    • Dispatched is identified as a regulator of sterol-modified Hedgehog release from cells. Dispatched displays structural homology to Patched, a receptor involved in both sequestration and transduction of Hedgehog signal. This suggests that a common mechanism may regulate sequestration and release of Hedgehog.

    • Expression of a BMP antagonist, Gremlin, by the distal limb mesenchyme is likely to mediate both maintenance and anterior extension of Fgf-4 expression in the AER. Using limb deformity mouse embryos, they show that expression of Gremlin is promoted indirectly by Shh signaling through Formin, an intracellular protein.

    • BMPs negatively regulate structure and function of the limb apical ectodermal ridge
    • Ectopic expression of either a BMP antagonist, Noggin, or a dominant negative form of the BMP receptor, Bmpr1b, upregulates Fgf-4 expression in the AER and maintains AER structure past the stage where regression normally begins. In addition, inhibition of signaling appears to block chondrogenesis and downregulate distally expressed genes, suggesting multiple roles for BMPs in limb development. However, Noggin is not expressed appropriately to mediate the early actions of BMP signaling.

    • ••]. Gremlin is expressed appropriately during early limb development. Viral-mediated expression of Gremlin results in defects similar to those described for Noggin. The authors also present evidence for a second antagonistic interaction in the early limb bud. Ectopic expression of Meis2, a homeodomain-containing protein expressed proximally, is shown to inhibit distal outgrowth.
