By comparing the time course of inhibitor effects, they report that rab action precedes SNARE action, producing a reversible association of vacuoles. SNARE proteins, on the other hand, mediate irreversible vacuole binding. Two unexpected results - first, that the rate of SNARE pairing does not match the rate of fusion and, second, that dissociation of the SNARE complex by NSF after docking does not affect subsequent fusion - suggest that SNAREs do not mediate membrane fusion but produce a stable state of association that is consistent with hemi-fusion. Post-docking fusion is sensitive to GTP-gamma-S and the phosphatase inhibitor microcystin LR, suggesting that multiple events may follow SNARE action
