dMi-2, a hunchback-interacting protein that functions in polycomb repression
-
Kehle J, Beuchle D, Treuheit S, Christen B, Kennison JA, Bienz M, Müller J: dMi-2, a hunchback-interacting protein that functions in polycomb repression. Science 1998, 282:1897-1900. dMi-2, a component of a histone deacetylase complex, is shown to bind the early repressor Hunchback (Hb), and to interact genetically with Polycomb (Pc) mutants. This provides the first connection between Hb-repression and Pc-repression and suggests a role histone deacetylation in mediating Pc-repression.

The dermatomyositis-specific autoantigen Mi2 is a component of a complex containing histone deacetylase and nucleosome remodeling activities
-
Zhang Y, LeRoy G, Seelig HP, Lane WS, Reinberg D: The dermatomyositis-specific autoantigen Mi2 is a component of a complex containing histone deacetylase and nucleosome remodeling activities. Cell 1998, 95:279-289. These papers [23•,24•] describe the purification of a new type of histone-deacetylase-activity-containing complex from Xenopus eggs and mammalian cells, that also has intrinsic ATP-dependent nucleosome-remodeling capacity. Moreover, the Mi-2 component is shown to be a member of the SWI2/SNF2 gene superfamily, suggesting a connection between different classes of chromatin-remodeling factors.

The Drosophila Polycomb group gene pleiohomeotic encodes a DNA binding protein with homology to the transcription factor YY1
-
Brown JL, Mucci D, Whiteley M, Dirksen ML, Kassis JA: The Drosophila Polycomb group gene pleiohomeotic encodes a DNA binding protein with homology to the transcription factor YY1. Mol Cell 1998, 1:1057-1064. The Drosophila Pc-G gene pleiohomeotic is shown to encode a sequence-specific DNA-binding protein that has homology to the pleiotropic mammalian transcriptional regulator YY1. This is the first example of a sequence-specific DNA binding Pc-G protein, which aids in understanding how Pc-G complexes interact with their target genes.

The Drosophila Fab-7 chromosomal element conveys epigenetic inheritance during mitosis and meiosis
-
Cavalli G, PaRo R: The Drosophila Fab-7 chromosomal element conveys epigenetic inheritance during mitosis and meiosis. Cell 1998, 93:505-518. This paper describes the use of a PRE flanking white and LacZ reporter genes, which can be released from Pc-G repression by a short pulse of the potent transcription activator GAL4. This allows the researcher to follow reporter gene expression in a temporal manner. A remarkable stability of active and inactive states is observed, persistent not only through mitosis but also, to some extent, through meiosis.

The oncogene and Polycomb-group gene bmi-1 regulates cell proliferation and senescence through the ink4a locus
-
Jacobs JJ, KIeboom K, Marino S, DePinho RA, Van Lohuizen M: The oncogene and Polycomb-group gene bmi-1 regulates cell proliferation and senescence through the ink4a locus. Nature 1999, 397:164-168. Functional and genetic experiments in mice place Bmi-1-interacting Pc-G proteins as upstream regulators of the tumor-suppressors p16 and p19Arf. These results provide a connection between Polycomb repression, cell-cycle regulation and cellular senescence, that is conserved in human primary cells.

The C-terminal SET domains of ALL-1 and trithorax interact with the INI1 and SNR1 proteins, components of the SWI/SNF complex
-
ROzenblatt-ROsen O, Rozovskaia T, Burakov D, Sedkov Y, Tillib S, Blechman J, Nakamura T, Croce CM, MAzo A, Canaani E: The C-terminal SET domains of ALL-1 and trithorax interact with the INI1 and SNR1 proteins, components of the SWI/SNF complex. Proc Natl Acad Sci USA 1998, 95:4152-4157. This paper demonstrates the direct interaction of a SWI/SNF component with the conserved SET domain, suggesting that this domain - found in trithorax and in proteins involved in position effect varigation - may act to recruit SWI/SNF to trithorax target genes.
