Morales CR, Wu XQ, Hecht NB: The DNA/RNA-binding protein, TB-RBP, moves from the nucleus to the cytoplasm and through intercellular bridges in male germ cells. Dev Biol 1998, 201:113-123. Cytoplasmic and nuclear fractions of mouse spermatocytes and spermatids are shown to bind, to different extents, to specific RNA and DNA probes. Immunoelectronmicroscopy confirms the shift of the protein from nucleus to cytoplasm and it is detected in intercellular bridges between spermatids.

The possibility that TRA2 itself regulates production of the full-length protein by a negative feedback mechanism is tested. Increased gene dosage of tra2 results in reduced levels of mRNA in which the third intron has been spliced. The effect of a transgene lacking the intron is more severe, and multiple copies reduce fertility progressively.

Dauwalder B, Mattox W: Analysis of the functional specificity of RS domains in vivo. EMBO J 1998, 17:6049-6060. This is one of a number of papers published in the past year or so reporting investigations of the contributions of individual RS domains to SR protein activities. Various RS domains were substituted for the second RS domain of TRA2 in transgenic flies. Female somatic differentiation, male fertility and subnuclear localization in spermatocytes were restored to varying degrees. Male fertility and TRA2 autoregulation were most sensitive and disrupted even by a carboxy-terminal epitope fusion with intact TRA2.

EMBO J 1999, 18:258-269. 3′ UTR sequences regulating translation of tra-2 are here used as bait in a three-hybrid screen. One clone shows specific binding and is found to encode GLD-1. Recombinant GLD-1 is shown to bind specifically and to inhibit translation of tra-2 in a yeast extract. Somatic expression of transgenes is used to show that GLD-1 inhibits translation in vivo of a lacZ reporter with tra-2 3′ UTR sequences.
