    • This manuscript provides evidence that T cell costimulation via CD28 receptors enhances the signaling environment of the engaged TCR. This implies that what had been previously thought of as two distinct stimulatory pathways actually converge at the membrane level.

    • Extrathymic T cell deletion and allogeneic stem cell engraftment induced with costimulatory blockade is followed by central T cell tolerance
    • This manuscript demonstrates that costimulation blockade combined with donor bone marrow transplantation is a very powerful strategy for achieving transplantation tolerance to solid organs. More importantly, the findings raise the possibility that costimulation blockade may obviate the need for depleting some of the host's bone marrow cells with toxic irradiation - traditionally required for successful engraftment of donor bone marrow cells.
