    • Cytomechanics of cadherin-mediated cell-cell adhesion
    • This is an excellent review on the subject of cadherin-mediated cell-cell adhesion. The authors also emphasise cadherin clustering and interactions with the actin cytoskeleton but more emphasis is placed on the regulation of the formation of cadherin dimers and proteins that mediate cadherin's lateral clustering and binding to actin filaments. Cytomechanical arguments have been advanced regarding the bidirectional influences between cell-cell adhesion and the actin/myosin network.

    • An excellent review on an interesting discovery made by this group. • Strength of a weak bond connecting flexible polymer chains
    • The strength of weak bonds connecting polymer chains was theoretically analysed. The effect of a polymer linkage is to weaken the strength of the bond from its expected logarithmic dependence, as a result of the non-linear force loading characteristics of the polymer linkage. A re-analysis of recent titin unfolding experiments showed this effect nicely.

    • Using dynamic force spectroscopy to explore energy landscapes of receptor-ligand bonds
    • Accurate measurements of the adhesive failure of biotin-avidin and biotin-streptavidin bonds were performed over a range of six orders-of-magnitude in the force-loading rate. The strength was found to be logarithmically dependent on the loading rate. This spectrum of strengths revealed the size and the position of the major energy barriers along the dissociation reaction pathway, which correlates well with predictions made from the receptor structures.

    • Adhesive but not lateral E-cadherin complexes require calcium and catenins for their formation
    • Triton-X-100-soluble proteins from cultured cells producing endogenous and recombinant forms of E-cadherin were subjected to sucrose gradient fractionation. Three distinct E-cadherin complexes were found: (i) one cadherin linked to a dimer composed of either β-catenin and α-catenin or plakoglobin and α-catenin, (ii) laterally-linked cadherin-catenin complexes via dimerization of the extracellular domain of E-cadherin, and (iii) two cadherin-catenin complexes derived from two opposing cells. The third complex is biologically most important and its formation required both extracellular calcium and catenin binding to cadherin as well as the amino acid residues in the amino-terminal domain that are responsible for lateral dimerization.

    • When almost all of the cadherin cytoplasmic domain was removed (only 0 or 7 amino acids remaining in the juxtamembrane region), the molecule exhibited the ability to form dimers and induce cell aggregation (but not compaction). E-cadherin mutants with deletions of 31 or 71 carboxy-terminal amino acid residues (with 120 or 80 amino acids remaining in the juxtamembrane domain) have no ability to aggregate cells, suggesting that the juxtamembrane domain can prevent lateral dimerization.

    • IQGAP1, a target of Cdc42 and Rac1, was localised in the cell-cell adhesion sites, and was found to interact with E-cadherin and β-catenin in vitro. IQGAP1 induced the dissociation of α-catenin from a cadherin-catenin complex both in vitro and in vivo, and reduced cell-cell adhesive activity. It was proposed that IQGAP1 regulates cell adhesion by dissociating cadherin from the actin cytoskeleton.

It was concluded that the movements of the wild-type E-cadherin in the plasma membrane in transfected L cells are regulated via the cytoplasmic domain by tethering to actin filaments through catenin(s) and the corralling effect of the network of the membrane skeleton.

    • Regulation mechanism of the lateral diffusion of band 3 in erythrocyte membranes by the membrane skeleton
    • Mechanisms that regulate the movement of band 3 protein in erythrocyte ghosts were investigated at the single molecule level using single particle tracking with an enhanced time resolution (0.22 ms) and dragging by laser tweezers. The results indicated that the cytoplasmic portion of band 3 collides with the membrane skeleton, which causes temporal confinement of band 3 inside a membrane skeleton mesh of 110 nm in diameter for 350 ms on average. Band 3 molecules hop to adjacent meshes, which enables them to undergo macroscopic diffusion.
