In this paper, the authors show that it is possible to predict to a large extent the in vivo dosage of a human growth hormone transgene under the control of the rapamycin inducible system. This elegant analysis also points to some of the limitations of the system in its current form, such as the slow kinetics of downregulation of the expression upon rapamycin withdrawal.

of special interest. This study describes the synthesis of rapamycin analogues that still bind FKBP12 but have lost the ability to bind the wild-type FRB from hFRAP. Furthermore, it describes the identification of FRB mutants that can be bound by these compounds by a three hybrid yest assay. Unlike rapamycin, these compounds should not be immunosuppressive and should thus prove useful to induce expression of exogenous genes in vivo.
