This study demonstrates that domestic swine DNA contains multiple copies of a genome (pig endogenous retrovirus) that encodes C-type retrovirus. This type of virus can be produced by two porcine kidney cell lines, one of which (the PK-15 cell line) is shown to be able to infect cultured human (kidney 293 cell line), pig and mink cells. It is not known whether normal pig cells in vivo release pig endogenous retrovirus particles and whether such particles replicate in vivo in humans.

Numerous inflammatory cells were identified 18-19 months post-transplantation within the surviving graft site at autopsy of a patient (case 2) who received cyclosporin for 6 months post-transplantation. Prior to death, the patient had a sustained clinical improvement and a progressive increase in flurodopa uptake between 6 and 12 months after cessation of cyclosporin. The clinical implications of this inflammatory reaction on the long-term survival and function of the graft remains unclear. It is conceivable that it may be detrimental (e.g. slow rejection, limitation of reinnervation of host targets) or beneficial (e.g. increased expression of neurotrophins induced by inflammatory mediators).

This report demonstrates that, in 6-OHDA-lesioned rats, GDNF given intranigrally or intraventricularly reverses the apomorphine-induced rotational behavior and the decreased nigral (but not striatal) tyrosine hydroxylase activity/dopamine levels. These effects are suggested to be due in part to peptidergic markers changes (striatal dynorphine A increase) involving the direct basal ganglia pathway. It is suggested that intraventricular injections of GDNF (although less influential on the peptidergic markers than intranigral injections) may be useful for the treatment of Parkinson's disease.

    • •]). Interestingly, the grafted cells displayed no Lewy bodies, had appropriately no neuromelanin given their age, and expressed tyrosine hydroxylase mRNA comparable to normal nigral neurons. They induced no sprouting of host tyrosine hydroxylase immunoreactive fibers as seen with adrenal graft, suggesting that this effect is not as critical for clinical recovery as graft survival and reinnervation.
