This paper documents the different consequences of neonatal injection of the zona pellucida peptide pZP3 (in incomplete adjuvant) in male mice as a foreign peptide, and in female mice as a self peptide. Male mice produce a Th2-biased response and do not develop disease in their ovarian grafts upon adult challenge. Females produce a mixed Th1/Th2 response and develop AOD five weeks after injection, unless their ovaries are removed between days two and seven of life, whereupon their response is similar to that of the male mice. The latter finding suggests the possibility that self antigens have an impact on the immune system early in ontogeny.

Science 1997, 276:604-607. NFS mice with s/d mutation, thymectomized on day three of life, developed spontaneous fodrin. Interestingly, the salivary glands of the thymectomized mice had over-expression of α-fodrin in the duct epithelium; both sialoadenitis and autoantibody response were prevented by neonatal in vitro injection of recombinant α-fodrin. Over 95% of patients with primary and secondary Sjogren's disease also had serum antibody to α-fodrin.

This study uses a powerful tool, a combinatorial peptide library, to identify cross-reactive ligands for a human CD4+ T cell clone that recognizes the myelin basic protein (MBP) peptide amino acid 86-96. Sublibraries containing 220 11-mer peptides were used to dissect the antigen recognition pattern of the T cell clones. This method easily identified stimulatory ligands from both human and microbial peptides, some being more potent than the MBP peptide, The general conclusion reached is that T cell recognition of cognate antigen is degenerate, and mimicry can occur without necessarily sharing common residues.

This paper, along with [27], uses the zona pellucida 3 (ZP3) peplide induced autoimmune ovarian disease (AOD) model to elucidate the following properties of autoantibody diversification: the induction of autoantibody production by the ZP3 T cell epitope or its mimic; the recognition of the native ZP3 B cell epitope by, and the functionality of, the diversified autoantibodies; the requirement of endogenous antigen from normal ovaries for autoantibody induction; and, the rapid kinetics of the autoantibody response. The findings indicate that the specificity of the autoantibodies may not mirror the antigen that initiates an autoimmune disease, and that B cell tolerance to self antigens depends mainly on the lack of T cell help.
