J Exp Med 1996, 183:1093-1104. Using the LCMV model, positive selection of a defined transgenic TCR is shown to occur in the presence of an agonist ligand, A4Y. Importantly, these T cells which have matured in the presence A4Y, no longer respond Io A4Y, but proliferate with the addition of a high-affinity agonist. This demonstrates that the T cell may adjust the resting threshold by interactions during positive selection.

Using transgenic mice expressing class II molecules with a covalently bound single peptide, in the absence of other class II molecules and the li, this study demonstrates that a single peptide-class II molecule is capable of selecting T cells with a diverse Vβ repertoire. It also shows that a significant amount of negative selection occurs in the thymus as a result of class II molecules presenting a variety of endogenous peptides.

Out of three different transgenic TCRs only one was positively selected, suggesting that the peptides required for selection of the transgenic TCR were not associated with class II in li-deficient mice. Interestingly, the response to one of the antigenic ligands for the transgenic TCR was also absent in li-deficient mice.
