J Clin Invest 1996, 97:2499-2508. In rats with mesangial proliferative nephritis induced by injecting anti-Thy-1 the expression of moesin and radixin is increased in infiltrating macrophages and in activated, alpha smooth muscle actin-positive mesangial cells in areas of glomerular remodeling. The increases in these cytoskeletal proteins were inhibited by antibodies to PDGF, but not FGF, suggesting that PDGF-mediated upregulation of cytoskeletal proteins is important for cell migration.

Using in-situ reverse transcription, the glomerular cell sites of alpha 1(1V) collagen, fibronectin, laminin B1, angiotensinogen and TGF-β1 mRNA synthesis was determined at sequential periods following renal ablation. The study provides evidence that as a result of hemodynamic changes, injured or activated endothelium synthesizes angiotensinogen, triggering a cascade of TGF-β1, and matrix protein gene expression with resultant development of the segmental glomerular sclerotic lesion.

This study provides in-vivo evidence that acute elevation in blood pressure induced by either restraint or administration of Ang II or phenylephrine is accompanied with a parallel increase in mitogen activated protein kinase activity in arterial wall. Furthermore, induction in mitogen activated protein kinase activity was inhibited in presence of SNP, an NO donor which lowers blood pressure.
