J Lipid Res 1996, 37:1849-1861. The cornea accumulates cholesteryl ester-rich lipid particles larger than LDL and similar to lipid particles in the lipid core of human atherosclerotic lesions. As there are no macrophages in normal human corneas, this type of lipid particle is likely derived from fused LDL and not from lipid droplets released from foam cells.

Proposed role for arterial-wall sphingomyelinase in subendothelial retention and aggregation of atherogenic lipoproteins. J Clin Invest 1996, 98:1455-1464. This research group has previously shown that sphingomyelinase causes LDL to aggregate. Now it has been shown that release of ceramide from sphingomyelin mediates this aggregation. Further, the potential for such a mechanism to operate in atherosclerotic lesions was shown. It was determined that in comparison with plasma LDL, aggregated LDL isolated from lesions is enriched in ceramide. Also, incubation of LDL with strips of rabbit aorta caused some LDL to be retained in the strips, and only the retained LDL showed hydrolysis of sphingomyelin to ceramide.

J Biol Chem 1996, 271:28641-28646. This report shows that human monocyte-macrophages have the capacity to efflux cholesterol through an autocrine mechanism dependent on macrophage apoE secretion. Thus, monocyte-macrophages in atherosclerotic lesions have the potential to excrete cholesterol independently of plasma-derived HDL. The finding may explain why transgenic expression of apoE in macrophages shows an anti-atherogenic effect (see Shimano et al. J Clin Invest 1995, 95:469-476 and Bellosta et al. J Clin Invest 1995, 96:2170-2179).

This study demonstrates that mast cell-derived chymase can interfere with the cholesterol acceptor function of HDL by proteolyzing its apoA-l. It will be interesting to learn whether other lesion proteases besides chymase (e.g. metalloproteinases) show a similar anti-HDL effect.
