Drug addiction: The yin and yang of hedonic homeostasis
-
••] provide a timely review of the types of animal models that are now available to study the neurobiology of drug addiction. The mounting evidence for an important role played by the mesolimbic dopamine system in mediating the reinforcing actions of drugs of abuse is summarized. In addition, the reviews highlight recent results that implicate other brain regions, particularly the amygdala, in negative motivational aspects of drug addiction.

Demonstrates that chronic morphine administration selectively upregulates two forms of adenylyl cyclase in the locus coeruleus, along with the catalytic and type II regulatory subunits of PKA. By infusing antisense oligonucleotides to CREB directly into the locus coeruleus, which decreases CREB levels in this region, the authors show that the morphine-induced upregulation of adenylyl cyclase, but not of PKA, is dependent on CREB function. They relate these biochemical findings to electrophysiological adaptations in locus coeruleus neurons and to behavioral signs of somatic opiate withdrawal.

This paper and [16] provide direct evidence that the cAMP pathway in the nucleus accumbens modulates the reinforcing properties of stimulants. Self and Nestler [16], using the drug self-administration paradigm, provide evidence that inhibition of the cAMP pathway increases the reinforcing properties of cocaine and can induce relapse to cocaine-seeking behavior. In contrast, Kelley and Holahan [17•], using the conditioned reinforcement paradigm, provide evidence that activation of the cAMP pathway increases the reinforcing properties of stimulants. These paradoxical findings clearly indicate the need for further study, but also highlight the importance of utilizing several complementary behavioral methods to infer modulation of reinforcement mechanisms in laboratory animals.

Sensitization to morphine induced by viral-mediated gene transfer

Carlezon WA Jr, Boundy VA, Haile CN, Kalb RG, Neve R, Nestler EJ: Sensitization to morphine induced by viral-mediated gene transfer. Science 1997, 277:812-814. The authors use recombinant herpes simplex virus vectors to overexpress glutamate type 1 receptor (GluR1) or other AMPA glutamate receptor subunits specifically in the ventral tegmental area. They show that overexpression of GluR1 specifically results in sensitized locomotor and reinforcing responses to morphine. Importantly, viral infection is transient and not associated with detectable toxicity. The results support the hypothesis that drug-induced upregulation of GluR1 in this brain region is one mechanism of drug sensitization. They also support the use of viral-mediated gene transfer to establish the molecular mechanisms underlying the complex behaviors of addiction.

This paper describes the generation, by homologous recombination, of mice lacking the dopamine transporter. The mice show marked increases in base-line locomotor activity, but no additional increase in response to acute administration of cocaine or amphetamine. The results support the important role played by the dopamine transporter in mediating the acute locomotor-activating effects of these stimulants. More significantly, the mice provide a unique system in which to determine the extent to which other effects of the drugs, particularly reinforcement and craving, are mediated by distinct mechanisms (e.g. inhibition of the serotonin or norepinephrine transporters). The mice also provide a unique system in which to study the brain's long-term adaptations to primary defects in the regulation of dopaminergic neurotransmission.

The authors generated mice deficient in the major forms of CREB, α and δ, by homologous recombination and demonstrated that they exhibit attenuated behavioral signs of somatic opiate withdrawal. The results thereby support a role for CREB in mediating some of the long-term adaptations to chronic opiate exposure. It will be interesting to determine whether other responses to opiates, particularly motivational adaptations, are similarly attenuated. The mice also provide a model system in which to identify some of the target genes through which CREB produces these effects. One limitation of the mice is that they do not provide information on the specific brain regions involved in opiate action, as the CREB deficiency is expressed ubiquitously.

The apparent discrepancies in these findings could be attributable to the different drugs, drug treatment regimens (e.g. intermittent versus continuous), or brain regions involved. These studies highlight the need for further research to more precisely define the way in which CREB is regulated by chronic drug exposure and to determine the functional (electrophysiological, behavioral) consequences of such regulation.

This paper definitively identifies long-lasting Fos-like proteins induced in nucleus accumbens and related striatal regions in response to chronic cocaine or morphine administration as isoforms of ΔFosB, a truncated splice variant of FosB (see also [63,65]). The behavioral significance of drug-induction of this transcription factor is demonstrated by sensitized responses to the locomotor-activating and reinforcing properties of cocaine in fosB knockout mice. A major challenge for future investigations is to identify the specific target genes through which changes in levels of ΔFosB lead to altered functioning of nucleus accumbens and other striatal neurons and, eventually, to modulation of behavioral responses to drugs of abuse.
