    • Integrin-modulating therapy prevents fibrosis and autoimmunity in mouse models of scleroderma
    • PID: 24107997, In this paper, two kinds of fibrillin 1 gene-mutated mice were generated to determine the mechanisms by which fibrillin 1 mutations cause stiff skin syndrome (SSS) in patients. Both lines of mutant mice phenocopied SSS with increased collagen deposition in the skin, suggesting that they could be good models for evaluating therapeutic interventions. The authors also showed that mutations in the integrin binding domain of fibrillin 1 resulted in the upregulation of α5β1 and αvβ3 integrins in dermal cells, especially plasmacytoid dendritic cells, and that inhibition of TGF-β or αvβ3 or activation of β1 integrins was effective in inhibiting the mutant phenotype. These results thus identified three potential targets for therapeutic intervention in SSS patients and perhaps in patients with cutaneous fibrosis from scleroderma
