Jones Stimulation of angiotensin AT2 receptors by the non-peptide agonist, compound 21, evokes vasodepressor effects in conscious spontaneously hypertensive rats Br J Pharmacol 159 2010 709 716 This is the first study looking at the impact of direct AT2R-stimulation with C21 on vasodilation and blood-pressure regulation. The authors show that C21 evoked dose-dependent vasorelaxations in aortic and mesenteric vessels, but lowered blood pressure in SHR only under concomitant AT1R-blockade.

Direct angiotensin II type 2 receptor stimulation acts anti-inflammatory through epoxyeicosatrienoic acid and inhibition of nuclear factor kappaB

Bader Direct angiotensin II type 2 receptor stimulation acts anti-inflammatory through epoxyeicosatrienoic acid and inhibition of nuclear factor kappaB Hypertension 55 2009 924 931 This study confirms the specific, AT2R-agonistic property of C21 for example by showing the absence of effects in AT2R-deficient cells. It furthermore provides evidence for the anti-inflammatory effect of C21 in vitro and in vivo and describes phosphatase and CYP2C/2J activation as well as NF-κB inhibition as underlying mechanisms.
