Lrp5 controls bone formation by inhibiting serotonin synthesis in the duodenum
-
1B these authors demonstrate that gut-derived 5-HT is an inhibitor of osteoblast proliferation whose increased blood level causes the bone loss associated with Osteoporosis Pseudoglioma in patients and Lrp5-deficiency in mice. This is the frst study showing that Lrp5 acts on bone mass by regulating Tph1 expression in gut.

A serotonin-dependent mechanism explains the leptin regulation of bone mass, appetite, and energy expenditure
-
2C as well as neuron tracing experiments to demonstrate that leptin mediates its central effects on bone mass accrual and appetite by inhibiting 5-HT synthesis in brainstem neurons. This paper also shows that mice deficient in both Tph1 and Tph2 have, as the Tph2-null mice have, a low bone mass phenotype caused by decreased bone formation and increased bone resorption.

Using a microarray strategy this study also identifies CREB target genes that mediates brain 5-HT inhibition of sympathetic output.

This is the first proof-of-concept study showing that specifically inhibiting 5-HT production in gut via a pharmacologic approach could become an anabolic treatment for osteoporosis.
