    • Roles of Phosphatidylinositol 3-kinase in root hair growth
    • The authors investigate the role of PI-3P production in root hair growth. AtVPS34 is shown to be broadly expressed in A. thaliana, and homozygous T-DNA insertions in this gene result in gametophytic lethal mutants. Overexpression of a GFP-2xFYVE fusion which binds and sequesters PI-3P, or treatment with the PI-3K inhibitor, LY294002 rapidly inhibited root hair elongation. Interestingly, LY294002 treatment did not inhibit early stages of endocytosis as measured by uptake of the styryl dye, FM 1-43.

    • Induction of phosphatidylinositol 3-kinase-mediated endocytosis by salt stress leads to intracellular production of reactive oxygen species and salt tolerance
    • The authors show that salt stress rapidly induces PI-3K activity and that this is associated with production of reactive oxygen species (ROS). Inhibition of PI-3K and PI-4K activities by wortmannin treatment was associated with reduced ROS production, and this was rescued by treatment with exogenous PI-3P. These results suggest that PI-3P mediates production of ROS under salt stress conditions.

    • This paper describes a careful evaluation of the gametophytic lethal phenotype of an atvps34 T-DNA insertional mutant line. Gametophytic lethality was shown to occur primarily in pollen and was shown to occur at late stages of pollen development. Specifically defects in vacuolar reorganization after the first mitotic division in tetrad microspores, often led to pollen with reduced numbers of nuclei which showed dramatically reduced levels of pollen germination and pollen tube formation as compared with wild type.

    • This paper identifies PI-4Kβ1 as a RabA4b effector protein, which is recruited to tip-localized membranes in root hair cells. Loss of PI-4Kβ activity results in aberrant root hair growth, implicating an important role for PI-4P production in polarized trafficking in these cells.
