Kinesin superfamily protein 2A (KIF2A) functions in suppression of collateral branch extension
-
-/- mice, whose brains show several phenotypes including aberrant axonal branching owing to overextension of collateral branches, is presented. The results show that KIF2A regulates microtubule dynamics at the growth cone edge by depolymerizing microtubules and that this KIF has an important role in the suppression of collateral branch extension, which is essential for brain wiring.

A common mechanism for microtubule destabilizers - M type kinesins stabilize curling of the protofilament using the class-specific neck and loops
-
T. Ogawa, R. Nitta, Y. Okada, and N. Hirokawa A common mechanism for microtubule destabilizers - M type kinesins stabilize curling of the protofilament using the class-specific neck and loops Cell 116 2004 591 602 This study presents a structural model of the mechanisms of microtubule depolymerization by KIF2. According to the model, its curved conformation allows KIF2 to bind tightly only at the end of the microtubules. Here, the L8 loop closes the nucleotide-binding pocket and ATP hydrolysis takes place. The neck helix destabilizes the lateral interaction of the protofilament, and the L2 loop stabilizes the curved conformation of the interdimer groove. Tubulin dimers or oligomers are spontaneously released from the curved end of the protofilaments.

W.L. Lee, J.R. Oberle, and J.A. Cooper The role of the lissencephaly protein Pac1 during nuclear migration in budding yeast J Cell Biol 160 2003 355 364 The authors propose that Pac1, the yeast homolog of the human lissencephaly protein LIS1, targets dynein to the microtubule tips, where the dynein and Pac1 are offloaded to the cortex, resulting in activation of dynein and sliding of the microtubules along the cortex of the bud.

Microtubules provide directional cues for polarized axonal transport through interaction with kinesin motor head
-
T. Nakata, and N. Hirokawa Microtubules provide directional cues for polarized axonal transport through interaction with kinesin motor head J Cell Biol 162 2003 1045 1055 This study shows that KIF5 has a preference towards microtubules in the initial segment of the axons. In addition, it shows that the carriers transported preferentially to axons are composed of tubules and vesicles, which are highly motile with long processivity. However, those transported to the dendrites are vesicular in shape, less motile with short processivity and evenly distributed within the cell body.

Evidence in support of signaling endosome-based retrograde survival of sympathetic neurons
-
H. Ye, R. Kuruvilla, L.S. Zweifel, and D.D. Ginty Evidence in support of signaling endosome-based retrograde survival of sympathetic neurons Neuron 39 2003 57 68 This paper shows that the kinase activity of the nerve growth factor (NGF) receptor, TrkA, is required in both distal axons and cell bodies for sympathetic neuron survival. It also shows that NGF that is retrogradely transported in cell bodies is crucial for neuronal survival but not for distal axon growth, reinforcing the hypothesis that retrograde transport is necessary for sympathetic neuron survival.

Oh Mutant dynactin in motor neuron disease Nat Genet 33 2003 455 456 In this study, a single base-pair mutation in the p150 subunit of dynactin is identified in a family with a slowly progressive, autosomal dominant form of lower motor neuron disease without sensory symptoms, showing that dynactin-mediated transport dysfunction can lead to human motor neuron disease.
