    • N. Kasthuri, and Lichtman JW The role of neuronal identity in synaptic competition Nature 424 2003 426 430 This study uses double transgenic mice expressing the cyan and yellow spectral variants of GFP in small numbers of motor neurons to examine synapse elimination at the neuromuscular junction. By imaging the whole axonal arbors of competing neurons, the authors observe that the neuron with the smaller arborization usually wins such competitions for synaptic territory. This indicates that limited global resources can determine the outcome of local competitions between neurons.

The reporter, synapto-pHluorin, is a pH-sensitive GFP variant fused to a synaptic vesicle protein and shows decreased fluorescence in the acidic environment of the vesicle lumen. On vesicle fusion and exposure to the extracellular space, the fluorescence increases and is used here as an indicator of synaptic activity in the olfactory bulb.

The use of fluorescent fusion proteins to tag subcellular structures is a strategy that is likely to become more widely used for in vivo neuronal imaging in the future.
