The role of neuronal identity in synaptic competition
-
N. Kasthuri, and Lichtman JW The role of neuronal identity in synaptic competition Nature 424 2003 426 430 This study uses double transgenic mice expressing the cyan and yellow spectral variants of GFP in small numbers of motor neurons to examine synapse elimination at the neuromuscular junction. By imaging the whole axonal arbors of competing neurons, the authors observe that the neuron with the smaller arborization usually wins such competitions for synaptic territory. This indicates that limited global resources can determine the outcome of local competitions between neurons.

In vivo imaging of neuronal activity by targeted expression of a genetically encoded probe in the mouse
-
T. Bozza, J.P. McGann, P. Mombaerts, and M. Wachowiak In vivo imaging of neuronal activity by targeted expression of a genetically encoded probe in the mouse Neuron 42 2004 9 21 This paper reports the first successful use of a genetically encoded reporter of neuronal activity in a transgenic mouse. The reporter, synapto-pHluorin, is a pH-sensitive GFP variant fused to a synaptic vesicle protein and shows decreased fluorescence in the acidic environment of the vesicle lumen. On vesicle fusion and exposure to the extracellular space, the fluorescence increases and is used here as an indicator of synaptic activity in the olfactory bulb.

In vivo imaging of synapse formation on a growing dendritic arbor

C.M. Niell, M.P. Meyer, and S.J. Smith In vivo imaging of synapse formation on a growing dendritic arbor Nat Neurosci 7 2004 254 260 Using time-lapse imaging and expression of a fluorescent synaptic marker protein, the authors examine the role of synapse formation in the stabilization of filopodia during dendritic arborization in the zebrafish optic tectum. The use of fluorescent fusion proteins to tag subcellular structures is a strategy that is likely to become more widely used for in vivo neuronal imaging in the future.

Stoeckli Screening for gene function in chicken embryo using RNAi and electroporation Nat Biotechnol 21 2003 93 96 This study illustrates how in vivo electroporation combined with RNA interference can be used in a relatively high-throughput manner to identify functionally important factors from a list of candidate genes.

A general method for gene knockdown in mice by using lentiviral vectors expressing small interfering RNA
-
•], shows that lentiviral vectors can be used to 'knockdown' the expression of a gene by RNA interference in transgenic animals, potentially providing a more rapid method of analyzing gene function as compared with traditional gene knockout approaches.
