    • Selective photostimulation of genetically chARGed neurons
    • A conceptually important study that formulates the idea of genetically targeted (photo)stimulation as an enabling technology for the analysis of neural circuits and the functional reconstitution of neurally encoded information. The idea is reduced to practice with the first genetically encoded (photo)trigger of neuronal activity, a metabotropic system termed 'chARGe'. The name commemorates the three essential protein components - the fly photoreceptor proteins arrestin, rhodopsin, and G-alpha - and their effect on neurons that express them: these neurons 'get a charge' out of light (that is, they spike upon illumination).

    • Transmission of olfactory information between three populations of neurons in the antennal lobe of the fly
    • The first functional analysis of a neural circuit with a genetically encoded optical probe of neuronal activity. Synapto-pHluorin, the reporter of synaptic transmission, is expressed in olfactory receptor neurons, projection neurons, and local interneurons of the antennal lobe of Drosophila, and odor-evoked synaptic activity in the intact fly brain is imaged using two-photon microscopy. The concluding paragraph of the paper envisions a more prominent future role for the fly in systems neuroscience, now that its powerful genetics and behavioral analysis can be supplemented with optical recordings of neuronal activity.

This might perturb neuronal physiology and raise the threshold for detection, as only calcium transients massive enough to fill all four calcium binding sites on G-CaMP elicit signals.
