    • The order of expression of transcription factors directs hierarchical specification of hematopoietic lineages
    • lo eosinophil progenitor cells (EoPs) in the normal bone marrow give rise exclusively to eosinophils. EoPs significantly increase in number in response to helminth infection, suggesting that the EoP stage is physiologically involved in eosinophil production in vivo. Authors also demonstrated that the expression of GATA-2 instructs C/EBPα-expressing GMPs to commit exclusively into the eosinophil lineage, while it induces basophil and/or mast cell lineage commitment if C/EBPα is suppressed at the GMP stage. They propose that the order of expression of key transcription factors is critical for their interplay to selectively drive lineage specification programs.

    • International Mepolizumab Study Group. In this study, the effect of mepolizumab on patients with moderate persistent asthma was evaluated by multicenter, randomized, double-blind, placebo-controlled study. While the administration of mepolizumab was associated with a significant reduction in blood and sputum eosinophils, there were no statistically significant changes in any of the clinical end points including morning peak expiratory flow, daily β2-agonist use, symptom scores, exacerbation rates, and quality of life measures.

    • Mepolizumab HES Study Group. In this study, the effect of mepolizumab on HES patients negative for Fip1-like 1-platelet-derived growth factor receptor α fusion gene (FIP1L1-PDGFRA) was evaluated by an international, randomized, double-blind, placebo-controlled trial. The authors showed that mepolizumab treatment enabled clinically significant reductions in corticosteroid dose, and often corticosteroid discontinuation, in HES patients without major safety concerns.

    • The absence of fucose but not the presence of galactose or bisecting N-acetylglucosamine of human IgG1 complex-type oligosaccharides shows the critical role of enhancing antibody-dependent cellular cytotoxicity
    • In this study, authors showed that anti-hIL-5Rα humanized IgG1 produced by rat hybridoma YB2/0 cell lines exerted more than 50-fold higher ADCC using purified human peripheral blood mononuclear cells as effector than those produced by Chinese hamster ovary (CHO) cell lines. Low fucose (Fuc) content of complex-type oligosaccharides was characteristic in YB2/0-produced IgG1s compared with high Fuc content of CHO-produced IgG1s. These results indicate that low- or no-fucose antibody induces the most powerful ADCC activity, and provide important information to produce the effective therapeutic antibody.
