The microtiter wells were coated with yeast mannan, unlabelled Con A was then allowed to attach to the polysaccharide, and the lectin-charged plate was subsequently incubated with HRP-labelled Con A. After the standard washing and visualisation protocol, no incorporation of Con A-HRP was observed, which precludes any significant HRP-induced aggregation of the lectin.
