This paper describes the first stages in the humanization of the N-linked glycosylation pathway of Pichia pastoris. Following the elimination of endogenous hyper-mannosylation, combinatorial libraries of mannosidase I and GnTI are used to produce the first in vivo hybrid N-glycan structures in yeast.
