    • Use of combinatorial genetic libraries to humanize N-linked glycosylation in the yeast Pichia pastoris
    • This paper describes the first stages in the humanization of the N-linked glycosylation pathway of Pichia pastoris. Following the elimination of endogenous hyper-mannosylation, combinatorial libraries of mannosidase I and GnTI are used to produce the first in vivo hybrid N-glycan structures in yeast.

By introducing three enzymes required for the synthesis of CMP-sialic acid, a CMP-sialic acid transporter and a sialyltransferase, the production of sialylated recombinant erythropoietin was demonstrated in humanized P. pastoris.
