    • An inward-facing conformation of a putative metal-chelate-type ABC transporter
    • The crystal structure of the metal-chelate-type ABC importer HI1470/71 from H. influenzae revealed an architecture similar to that of BtuCD. However, HI1470/71 was trapped in an inward-facing conformation, with the translocation pathway exposed to the cytoplasm. Comparison with BtuCD revealed conformational changes that may be relevant in the 'alternating access and release' mechanism.

    • Structure of an ABC transporter in complex with its binding protein
    • The crystal structure of the molybdate/tungstate ABC importer ModBC from A. fulgidus was determined in complex with its cognate binding protein ModA. The observed transmembrane architecture is probably conserved in molybdate, sulfate, and phosphate transporters, and may even extend to other ABC importers that have a total of 10-12 transmembrane helices. The structure reveals a nucleotide-free state coupled to an inward-facing conformation, suggesting a common mechanism for ABC transporters (see also text).

The structure was determined with bound ADP but reflects the ATP-bound state, with an outward-facing conformation of the transmembrane domains.

    • The structure of the multidrug ABC transporter Sav1866 in complex with AMP-PNP, a non-hydrolyzable ATP analogue. The structure reflects a bona fide ATP-bound state, demonstrating that the original Sav1866 structure, determined with bound ADP, had indeed revealed the ATP-bound state.
