They found that conditional loss of PTEN in selected brain regions led to changes in dendritic and axonal growth and synapse number. These mice also developed macrocephaly and behavioral abnormalities that are similar to certain features of ASD.

The authors found that loss of TSC1 or TSC2 leads to increased soma size, decreased dendritic spine density and changes in synaptic strength. These results suggest that defects in dendritic morphology and synaptic function contribute to the neurological symptoms of tuberous sclerosis, a disorder that often presents with autism.
