Expression of HIV-1 genes in podocytes alone can lead to the full spectrum of HIV-1-associated nephropathy
-
Zhong J, Zuo Y, Fogo A, et al. Expression of HIV-1 genes in podocytes alone can lead to the full spectrum of HIV-1-associated nephropathy. Kidney Int 2005; 68:1048-1060. Details the findings that podocyte-specific expression of HIV-1 in a murine model is sufficient for the development of collapsing glomerulopathy. Pivotal study which establishes that infection of podocytes is essential for inducing the HIVAN phenotype.

HIV-1 Nef induces dedifferentiation of podocytes in vivo: A characteristic feature of HIVAN
-
Husain M, D'Agati VD, He JC, et al. HIV-1 Nef induces dedifferentiation of podocytes in vivo: a characteristic feature of HIVAN. AIDS 2005; 19:1975-1980. Describes a murine model of podocyte-specific expression of nef. The findings suggest that nef expression is sufficient to induce podocyte de-differentiation, expression of proliferation markers, and activation of signaling pathways leading to cell proliferation. Provides compelling in-vivo evidence that nef alters podocyte phenotype and is essential for causing the early molecular changes which characterize HIVAN.

Activation of PAK2 and its association with nef are conserved in murine cells, but are not sufficient to induce an AIDS-like disease in CD4C/HIV transgenic mice

J Biol Chem 2006; 281:6940-6954. Suggests a possible role for the interaction and activation of human p21-activated kinase 2 (PAK2) by nef which leads to AIDS-like disease in several target organs in transgenic mice. Identifies another possible signaling mechanism through which nef may exert its pathogenic effects in the kidney.
