    • An essential role for CoREST in nucleosomal histone 3 lysine 4 demethylation
    • •• ]. The authors show that LSD1, a nuclear amine oxidase homolog, is a histone H3 lysine 4 demethylase and its activity is regulated by associated protein co-factors. The demethylation process mediated by LSD1 is highly selective for H3-K4 mono- and di-methylation, but does not work for H3-K4 tri-methylation.

Gamblin Regulation of p53 activity through lysine methylation Nature 432 2004 353 360 The authors identify a novel mechanism of p53 regulation through lysine methylation by Set9 methyltransferase. In response to DNA damage, p53 is methylated and, hence, its stability is increased. The methylated p53 is in the nucleus and regulates p53 target genes.

    • I.H. Su, M.W. Dobenecker, E. Dickinson, M. Oser, A. Basavaraj, R. Marqueron, A. Viale, D. Reinberg, C. Wulfing, and A. Tarakhovsky Polycomb group protein ezh2 controls actin polymerization and cell signaling Cell 121 2005 425 436 In this study, the authors report the existence of a cytosolic Vav1-containing Ezh2 methyltransferase complex and its function in the regulation of actin polymerization-dependent processes such as antigen receptor signaling in T cells and PDGF-induced dorsal circular ruffle formation in fibroblasts. The cytosolic Ezh2 controls actin polymerization through Rho GTPase in a methylation-dependent manner. The authors suggest cytosolic lysine methylation as a novel post-translational modification in signal transduction.

    • T.L. Cha, B.P. Zhou, W. Xia, Y. Wu, C.C. Yang, C.T. Chen, B. Ping, A.P. Otte, and M.C. Hung Akt-mediated phosphorylation of EZH2 suppresses methylation of lysine 27 in histone H3 Science 310 2005 306 310 The authors show that Akt interacts with and phosphorylates Ezh2. Akt-mediated Ezh2 methylation does not change its interaction with co-factors Eed and Suz12 but reduces its affinity towards histone H3 and, hence, the methylation of H3-K27. The authors speculate on the role of Ezh2 phosphorylation in oncogenesis through de-repression of Ezh2's target genes.
