Genotypes in eight candidate genes (COL1A1 and COL1A2, osteocalcin, osteonectin, osteopontin, VDR, ER, and AR) were examined in relation to bone density in 428 healthy non-Hispanic white children (228 girls and 200 boys aged 4.5-6.5 years). BMD/BMC measures were most consistently associated with COL1A2 and osteocalcin genotypes, less consistently with osteonectin, osteopontin, and VDR polymorphisms and not at all with COL1A1 (Rsal and Sp1), VDR (Bsml), or ER polymorphisms (Pvull, Xbal, TA). There were also gene-by-gene interaction effects for ER with VDR and osteocalcin.
