J Am Chem Soc 1999, in press. Conformationally rigidified β-peptide oligomers up to hexamer length were derived from optically active trans-2-aminocyclohexanecarboxylic acid subunits. X-ray crystallography reveals a helical structure that is stabilized by a network of 14-membered hydrogen-bonded rings between the backbone carbonyl and NH groups. Circular dichroism studies obtained in methanol suggest that this secondary structure is stable in solution. Strategies for the design of tertiary structures stabilized by the patterning of hydrophobic and polar residues are presented.

The solution structures of water-soluble β-peptide hexamers were studied. These polycationic compounds are synthesized with specific sequences of diverse cyclohexyl and acyclic residues. A high-resolution structure, the first for an unnatural oligomer in aqueous solution, was obtained by 2D NMR spectroscopy and reveals a '14 helix'.

A stable secondary structure for the γ-peptide was characterized by 2D NMR spectroscopy in organic solvents. A right-handed helical structure is described with a pitch of 5 Å and defined by the formation of 14-membered hydrogen-bonded rings along the backbone.

The conformation is defined by 14-membered hydrogen-bonded rings, creating right-handed helices with a pitch of approximately 5 Å. The structure appears to be stable up to 323K.

Proc Natl Acad Sci USA 1998, 95:4303-4308. Sequence-specific N-substituted glycine polymers (peptoids) up to 36 residues in length were synthesized using a robotic solid-phase protocol. A diverse set of chiral sidechains were incorporated, including aliphatic, aromatic, heterocyclic, cationic and anionic groups. CD spectra indicative of helical secondary structures are observed in both aqueous and organic solvents at temperatures up to 65°C. This structure is stable even at short chain lengths (five residues), despite the inability of these molecules to form backbone hydrogen bonds.

The major species present is a right-handed helix with repeating cis-amide bonds. The helix has a periodicity of three residues per turn and a pitch of approximately 6 Å. Cis→trans isomerization is found to be a source of conformational heterogeneity. These results are consistent with previously reported spectroscopic and modeling data.
