Retrovirus envelope protein complex structure in situ studied by cryo-electron tomography

F. Forster, O. Medalia, N. Zauberman, W. Baumeister, and D. Fass Retrovirus envelope protein complex structure in situ studied by cryo-electron tomography Proc Natl Acad Sci USA 102 2005 4729 4734 This paper demonstrates that 3D images of Moloney murine leukemia retrovirus particles can be obtained at a resolution high enough to identify the individual glycoprotein trimers that protrude from the surface of the virus. The authors show that the coherent averaging of a number of individual trimers computationally extracted from the tomogram results in a density map in which individual domains of the complex can resolved.

Serial block-face scanning electron microscopy to reconstruct three-dimensional tissue nanostructure
-
W. Denk, and H. Horstmann Serial block-face scanning electron microscopy to reconstruct three-dimensional tissue nanostructure PLoS Biol 2 2004 e329 A strategy is presented for the reconstruction of large volumes of tissue by automated imaging of the face of a plastic-embedded tissue block that is being sectioned with an ultra-microtome situated within the chamber of a scanning electron microscope. It is shown that images from the block can be obtained at intervals of 50-70 nm, which suggests that it should be possible to reconstruct volumes that span several tens of microns.
