The major interest of the paper is the positioning of polymerase λ3 within the core shell of the virion.

Visualization of a-helical features in a density map constructed using 9 molecular images of the 1.8 MDa icosahedral core of pyruvate dehydrogenase

M.J. Borgnia, D. Shi, P. Zhang, and J.L.S. Milne Visualization of a-helical features in a density map constructed using 9 molecular images of the 1.8 MDa icosahedral core of pyruvate dehydrogenase J Struct Biol 147 2004 136 145 The authors demonstrate interesting statistical data on the EM structural analysis of a highly symmetrical molecule and comparison with the known crystallographic structure. Results shown in the paper are surprising and draw attention to problems related to analysis of noisy images of particles with structural variations.

Optimal determination of particle orientation, absolute hand, and contrast loss in single-particle electron cryomicroscopy
-
P.B. Rosenthal, and R. Henderson Optimal determination of particle orientation, absolute hand, and contrast loss in single-particle electron cryomicroscopy J Mol Biol 333 2003 721 745 The authors formulate a new method for validation and optimisation of single-particle reconstructions, and analyse the resolution criteria. Image pairs are collected with a small difference in tilt angle, and a previously determined reconstruction is used to test the match between predicted and observed tilt views. The procedure avoids the major technical difficulties of high tilt and does not require a large data set for testing. It can be used to determine the hand of the structure.
