Egr-1 binding to a recognition sequence in the TIMP-1 promoter was demonstrated. The same DNA sequence is also recognized by the transcription factor Sp-1. In addition, RASF expressing Egr-1 at high levels were found to express increased levels of TIMP-2 and TIMP-3 mRNA.

The study demonstrates that invasion of cartilage is mediated by activated RASF accompanied by increased expression of adhesion molecules, matrix-degrading enzymes, but does not depend on cellular proliferation, as demonstrated by using SV40 immortalized RASF in comparison with nontransformed RASF.

When compared with MTX, significantly higher amounts of albumin accumulate in inflamed paws. The protein is metabolized by human RASF in vitro and in vivo. MTX-HSA was significantly more effective in onset suppression of arthritis in mice than was MTX.
