A plot of similarity of interaction, as measured by studying known three-dimensional structures, reveals how interactions diverge as a function of sequence similarity. Binding modes of interacting protein pairs that share above 30% sequence identity are generally similar. Below this cutoff lies a twilight zone, where the modes of interaction may or may not be similar.
