This study in a human cortical-collecting-duct cell line shows that activators of PPARγ enhance the activity of SGK1 through direct upregulation of SGK1 mRNA expression. This increases cell-surface expression of ENaC after 4 h, and ENaC mRNA and protein expression after 24 h. This suggests that SGK1 and ENaC are targets of exogenous and endogenous PPARγ activators.

Experiments in Fischer rat thyroid cells and COS-7 cells show that cAMP inhibits Nedd4-2 by decreasing its binding to ENaC. Three Nedd4-2 residues have to be phosphorylated by PKA for cAMP to inhibit Nedd4-2. These are the same residues phosphorylated by SGK1. Activation of either the SGK1 or cAMP pathway decreases the stimulation of ENaC by the other pathway. This indicates that Nedd4-2 integrates effects of cAMP and SGK1.
