This paper demonstrates that NF-κB activation in a human T cell line induced by a variety of diverse stimuli could be inhibited by the antioxidant N-acetyl-I-cysteine. The authors suggest that all the agents tested, whilst originally thought to activate NF-κB by distinct intracellular pathways, may in fact share common mechanisms and suggest that reactive oxygen intermediates serve as the widely used second messenger in the activation of NF-κB

In this review the authors critically examine the evidence for the universal role of oxidative stress in NF-κB activation. They find that the majority of the data in support of the oxidative stress model of NF-κB activation are specific to a stimulus in a cell line. This paper concludes that the evidence does not point to a central role for reactive oxygen species (ROS) and suggests that it is now important when proposing a role of ROS in a pathway to identify the particular kinase or adapter molecule that is redox modulated. These authors stress the need to treat individual pathways of NF-κB activation as distinct and suggest that the role of oxidative stress is the exception rather than the rule.

Nucleic Acids Res 1992; 20:3821-3830. This is one of the first publications identifying the necessity of the conserved cysteine residue in the DNA binding loop of NF-κB p50. The authors use purified NF-κB proteins and site-directed mutagenesis to identify cysteine 62 of p50 as being the important residue. Substitution of this cysteine for a serine decreased the DNA activity and rendered the protein insensitive to thiol modifying agents. The authors were also able to conclude that thioredoxin stimulates DNA binding of NF-κB by reduction of a disulphide bond involving cysteine 62.
