-
Marsh JB, Welty FK, Lichtenstein AH, et al. Apolipoprotein B metabolism in humans: studies with staple isotope-labeled amino acid precursors. Atherosclerosis 2002; 162:227-244. This is a comprehensive review of apoB kinetic studies employing stable isotope tracers. Included in this review are discussions of experimental methodologies and methods of data analysis. In addition, studies examining the effects of aging, diet, dyslipidemias, hormones and statins upon apoB metabolism are presented.

Apolipoprotein B100 kinetics in visceral obesity: Associations with plasma apolipoprotein CIII concentrations
-
Chan D, Watts GF, Redgrave TG, et al. Apolipoprotein B100 kinetics in visceral obesity: associations with plasma apolipoprotein CIII concentrations. Metabolism 2002; 51:1041-1046. Plasma lipid and lipoprotein abnormalities observed in visceral obesity are due to a combination of over secretion and impaired catabolism. This study demonstrated a strong positive association between VLDL apoB secretion and apoCIII concentration. This association remained after adjusting for insulin resistance, free fatty acids and cholesterol synthesis. VLDL apoB FCR was inversely associated with apoCIII.

3-leucine. A compartment model was used for the analysis of the tracer data. Fish oil reduced VLDL apoB secretion and the conversion of VLDL to LDL. Atorvastatin increased VLDL, IDL and LDL fractional catabolic rates with no effect of apoB secretion. Combined therapy produced additive affects on plasma triglycerides, HDL cholesterol, and VLDL apoB.

Body weight modulates cholesterol metabolism in non-insulin dependent type 2 diabetes
-
Simonen P, Gylling H, Miettinen TA. Body weight modulates cholesterol metabolism in non-insulin dependent type 2 diabetes. Obes Res 2002; 10:328-335. Following on from their previous studies [54] suggesting that low cholesterol absorption and high synthesis may be part of the metabolic syndrome, this study reports on the association between insulin resistance, cholesterol synthesis and cholesterol absorption. With increasing insulin resistance, cholesterol synthesis rises and the absorption of cholesterol falls.

Additive effect of plant-sterol ester margarine and cerivastatin in lowering low-density lipoprotein cholesterol in primary hypercholesterolemia
-
Simons LA. Additive effect of plant-sterol ester margarine and cerivastatin in lowering low-density lipoprotein cholesterol in primary hypercholesterolemia. Am J Cardiol 2002; 90:737-740. A multicentre, randomized trial with four parallel treatment arms on the effects of plant sterols (2 g) and cerivastatin (400 μg) treatment in hypercholesterolemic subjects. Sterol and statin reduced LDL cholesterol concentrations 8 and 32%, respectively. Sterol plus statin treatment demonstrated additive effects with a 39% reduction in LDL cholesterol.

Inheritance of cholesterol metabolism of probands with high or low cholesterol absorption
-
Gylling H, Miettinen TA. Inheritance of cholesterol metabolism of probands with high or low cholesterol absorption. J Lipid Res 2002; 43:1472-1476. Cholesterol absorption studied in siblings of hypercholesterolemic patients revealed a significant heritable component. Cholesterol absorption was measured as the ratio of plant sterols to plasma cholesterol. Cholesterol synthesis was also estimated from the measurement of plasma cholesterol precursors.
