    • Structural analyses of DNA recognition by the AML1/Runx-1 Runt domain and its allosteric control by CBFβ
    • Crystal structures of the Runx1-CBFβ-DNA and Runx1-DNA complexes are presented. The fine structural changes of the Runx1-DNA complex upon CBFβ binding, together with the results of extensive functional assays using various mutants, indicate the possible mechanism for the allosteric regulation of the Runx1-DNA interaction by CBFβ. Also, the leukemia-inducing mutation points are mapped on the complex structure

    • Mechanism of c-Myb-C/EBPβ cooperation from separated sites on a promoter
    • c-Myb cooperates with C/EBP family members to regulate gene expression in myeloid differentiation, whereas AMV v-Myb, an oncogenic mutant, does not. Crystal structures of the c-Myb(R1R2R3)-C/EBPβ(bZip)-DNA and AMV v-Myb(truncated R1R2R3)-C/EBPβ(bZip)-DNA complexes, and AFM images of these complexes using the native mim-1 promoter DNA are presented. Interactions between c-Myb R2 and the leucine-zipper part of C/EBPβ are accompanied by DNA looping in the c-Myb-C/EBPβ-DNA complex. In contrast, no Myb-C/EBPβ interaction and no DNA looping are found in the AMV v-Myb-C/EBPβ-DNA complex. These results are consistent with those obtained from Myb-C/EBPβ interaction experiments in solution using the GST pull-down assay and from Myb-C/EBPβ trans-activational cooperativity experiments using the luciferase assay
