This article provides evidence that cationic proteins released after cross-linking of PMN-expressed CD11b/CD18 stimulate alterations in endothelial barrier function. Furthermore, this article provides conclusive evidence that adherence-dependent CD11b/CD18 signaling and PMN-induced alteration in endothelial permeability are not dependent upon PMN transmigration.

Transmigration occurred through pre-existing gaps or after the formation of transient gaps in VE-cadherin junctional distribution. Gaps closed up after migration of PMNs.

This article describes PMN-stimulated alterations to epithelial barrier function and demonstrates that enhanced permeability occurred independently of transmigration but required PMN-epithelial contact. PMNs stimulated phosphorylation of endothelial MLC, indicating contraction of the actin-myosin ring and phosphorylation of junctional proteins on tyrosine residues.
