    • Dendritic spine geometry is critical for AMPA receptor expression in hippocampal CA1 pyramidal neurons
    • Using focal two-photon photolysis of a new caged glutamate compound (MNI-glutamate), the authors mapped glutamate sensitivities of individual dendritic spines of hippocampal CA1 neurons. They find that the expression of functional AMPA receptors correlates with spine geometry. Whereas thick mushroom spines showed high glutamate sensitivity, thin spines and filopodia minimally expressed functional AMPA receptors, suggesting the presence of silent synapses on these spines. Using fluctuation analysis, the number of functional AMPA receptor channels in mushroom spines was estimated to be in the range of 50 to 150.

    • Two-photon microscopy in brain tissue: Parameters influencing the imaging depth
    • Here, the authors discuss the dependence of depth penetration on the angular and area acceptance of detection optics. They provide some very useful measurements of scattering parameters of brain slices and intact neocortex. High NA, low-magnification objectives (e.g. the 20x/0.95 NA water immersion lens from Olympus) are shown to be particularly well suited for deep imaging in scattering tissue. This paper finds that depth penetration with such an objective increases by more than 100 μm compared to standard objectives.

    • Fast 100-nm resolution three-dimensional microscope reveals structural plasticity of mitochondria in live yeast
    • In this paper, a multifocal multiphoton 4Pi-confocal microscope, combining fast three-dimensional imaging with axial superresolution, is presented. For the first time, an equilateral resolution of 100 nm is achieved in living cells. This enabled the morphological analysis of GFP-labeled mitochondrial networks in yeast growing on different carbon sources.

    • Christie et al. provide the first demonstration of long-term imaging with high-resolution in the neocortex of living animals. Growth and stability of senile plaques (β-amyloid peptide deposits) in a mouse model of Alzheimer's disease were monitored using in vivo multiphoton microscopy. Individual plaques could be followed over weeks and months by repeated imaging through a thinned bone window. Most plaques were found not to change size after their initial formation. This is evidence for the 'dynamic feedback' model of plaque formation. The thinned-skull preparation is minimally invasive and should be useful for long-term imaging of neuronal structure and activity.
