Cell 1999, 97:189-198. Mice that lacked gp130 receptor only on their cardiac myocytes developed LV chamber dilation and heart failure after being subjected to a hemodynamic stress. Widespread apoptosis was seen in the myocardium of these gp130-deficient mice. This suggests that gp130-signals normally function to prevent this apoptosis during cardiac hypertrophy, providing an as-yet unknown survival signal.

Although many studies have suggested that Gq-signaling is important for the development of cardiac hypertrophy and possibly CHF, this is the first study to show that they are also necessary. The authors created transgenic mice that overexpress a peptide corresponding to the carboxy-terminus of Gαq specifically in cardiac myocytes. This inhibitory peptide was sufficient to attenuate the normal cardiac hypertrophy that develops after aortic banding. These studies suggest that this might be a useful therapeutic strategy in humans.

Cell 1999, 98:465-474. Deletion of delta-sarcoglycan resulted in the development of abnormalities in the coronary microvasculature and subsequently cardiomyopathy. This could be prevented completely by treatment of these mice with vascular smooth muscle relaxants. This is particularly important since abnormalities in the coronary vasculature have been reported in more common forms of DCM and suggest a novel mechanism for this finding.

The growing number of transgenic mice that model human disease or have perturbations in pathways potentially important for the development of heart failure now allow the direct testing of mechanistic hypotheses in vivo. These articles described the use of an inhibitor of βARK1 or phospholamban-deficient mice to rescue the phenotype in a genetic model of CHF in mice. It is likely that the identification of these novel therapeutic pathways will lead to the development of pharmacologic inhibitors that will change the way we treat patients with CHF.
